Recent advances in stem cell research made possible to generate pluripotent cells and direct differentiation that could indeed translate into effective therapies for certain human disorders. However, access to unlimited numbers of specific cell types for delivery and guide differentiation for reconstructions of tissue or organs on demand has been a long-standing goal in tissue engineering and regenerative medicine. Cellular differentiation and development are a large part epigenetic events that are heritable changes in gene activity that are not caused by changes in the DNA sequence. A global picture of DNA methylation and chromatin states in pluripotent cells and their changes during differentiation has been the focus of most published studies. It is important to understand the epigenetic impact on tissue engineering.
